Changes Associated with the Appearance of Mature Sugar Cataracts.

چکیده
Analyses for lens sugar alcohols, fructose, glucose, ATP, dry weight, hydration, and extracellular space were made before and after the appearance of mature diabetic and galactose cataracts. The appearance of mature cataracts in galactose-fed rats is associated with a marked and sudden decrease in the level of lens sugar alcohol, ATP, and dry weight, and an increase in lens hydration and extracellular space, as determined with radioactive sorbitol. Similarly, in the lenses from diabetic rats there is a sudden decrease in lens sugar alcohol, fructose, ATP, and dry weight, and an increase in lens glucose, hydration, and extracellular space. The results are interpreted as indicating that the appearance of mature cataracts is associated ivith the disruption of the fiber membranes of the lens.

Physiological and Optical Alterations Precede the Appearance of Cataracts in Cx46fs380 Mice
Purpose Cx46fs380 mice model a human autosomal-dominant cataract caused by a mutant lens connexin46, Cx46. Lenses from Cx46fs380 mice develop cataracts that are first observed at ∼2 months in homozygotes and at ≥4 months in heterozygotes. The present studies were conducted to determine whether Cx46fs380 mouse lenses exhibited abnormalities before there are detectable cataracts. Methods Lenses...
متن کاملGalactose cataract prevention with sorbinil, an aldose reductase inhibitor: a light microscopic study.
Cataract formation in galactosemic rats was studied by ophthalmoscopy, slit-lamp biomicroscopy, and by light microscopy using plastic embedding with methacrylate. Untreated rats developed nuclear cataracts by 14 days and mature cataracts by 21 days. However, rats treated with the aldose reductase inhibitor sorbinil did not develop any cataractous change for up to 8 months of 50% galactose feedi...

متن کاملScopoletin Inhibits Rat Aldose Reductase Activity and Cataractogenesis in Galactose-Fed Rats
Cataracts are a major cause of human blindness. Aldose reductase (AR) is an important rate-limiting enzyme that contributes to cataract induction in diabetic patients. Scopoletin is the main bioactive constituent of flower buds from Magnolia fargesii and is known to inhibit AR activity. To assess scopoletin's ability to mitigate sugar cataract formation in vivo, we studied its effects in a rat ...
